Well, you SHOULD be able to... and technically yes you can change your homepage, and delete the file. But there's some sort of virus, spyware or other third party software installed on your computer that keeps reproducing the .dll file and implementing the unwanted changes.

OK, a quick google for "IE Homepage Rewrite" came up with a few things.

I think the first thing to do is to uninstall IE, and then do a complete reinstall/upgrade.
The second link also contains a few anti-hijack software. If the reinstall didn't work, try them. Apparently, they should work, but depending on how new this malware is, they might not.
Try googling the web for "IE Homepage Rewrite" (or something similar), most of the fixes is rather technical, and requires you to edit your registry, so it is best that you read them yourself. I wouldn't feel comfortable guiding you through some registry hacks etc.
This is a ugly workaround, but if all else fails, try to create a new shortcut on the desktop and use this for the command line:
"C:\Program Files\Internet Explorer\IEXPLORE.EXE" -nohome
(assuming your iexplore.exe lives at that path). This will start IE with no homepage. (not really what you want, I know).
If you can't get it to work, post back here and I'll see if I can help in some way.
Lastly, consider changing your browser. Try out a few: Opera, Mozilla etc. Not only are they not as susceptable (sp?) to malicious exploits, they also enhance your browsing experience with a range of features that is lacking from IE (*cough*cough* mouse-gestures *cough*cough*)
EDIT: please don't refer to people that write malicious code (and other stuff that is malicious) as hackers. The correct term is crackers. This is like refering to footbag as hackysack (its even worse).

Firefox is put out by the Mozilla group, and is as Mozilla product. I'm not totally sure if Firefox is included with the Mozilla Suite package or not, as I've never installed it (the Mozilla Suite).
